My office is giving a USB drive with the same contents to 50 in-service participants at a training next week. Is it possible to create a rule that whenever I insert a USB Drive, a folder automatically copies from my Mac to the USB drive?

Hazel doesn't really have a "copy from" action. You can use a script or Automator action to do that part though.
